| Türkçe Özet | Bu çalışmada depresif hastaların eritrosit membran yağ asidi kompozisyonu araştırılmıştır. Bu amaçla; ilaç kullanmayan 30 kişilik depresif hasta grubundan ve 30 sağlıklı gönüllü kontrol grubundan kan örnekleri alınmıştır. Kan numuneleri alındıktan sonra eritrosit paketi hazırlanarak eritrosit sayımı yapılmıştır. Sayımı yapılan paketten Rose-Oklander metoduyla lipidler ekstrakte edilmiştir. Bu ekstraksiyon kullanılarak iki yönlü İnce Tabaka Kromatografisi (TLC) ile fosfolipidler belirlenmiştir. Belirlenen bu fosfolipit fraksiyonlarında yağ asidi metil esterleri oluşturularak gaz kromotografisinde cins ve miktarları tespit edilmiştir. Yapılan çalışmalarda n-3, n-6 seviyeleri ile n-6/n-3,ALA/EPA, EPA/DHA oranları depresyonla ilgili kriterler olarak belirtilmiştir. Literatürlerde genel olarak n-3 ve n-6 yağ asitleri depresif hasta grubunda düşük bulunmuştur. Bizim çalışmamızda da bu yağ asitleri düşük olarak gözlemlenmiştir. EPA, ALA, DHA miktarları ise farklı çalışmalarda farklı olarak bulunmuştur. Bazı çalışmalarda bu yağ asitleri miktarları düşük bazı çalışmalarda ise yüksek olarak tespit edilmiştir. Bizim çalışmamızda ise AA'da bir azalma DHA'da ise anlamlı olmayan bir artış olduğu tespit edilmiştir. Bu da ALA'nın siklooksijenazla DHA'ya çevrildiğini göstermektedir. |
| İlgilizce Özet | In this research, the erythrocyte membran fatty acid composition of depressive patients were investigated. To pursue this goal, blood samples were taken from 30 depressive patients who don?t use medicine and 30 volunteer, control subject. After this, erythrocyte packet were preparem and erytrocyte enumeration was done. Lipids were extracted from the enumerated packets using Rose-Oklander methodology. By using this extraction two-sides TLC (thin layer chromotography) and phospholipids were determined. Constituting fatty acids methyl esters in these phospholipids fractions, types and quantities in gas chromotograpy were found. In other studies, the levels of n-3, n-6 and the ratios of n-6/n-3, ALA/EPA, EPA/DHA were indicated as criteria related to the depression. In literatures, n-3 and n-6 fatty acids were generally found in low levels in the group of depressive patients. In our study, these fatty acids were also found in low levels. The amounts of EPA, ALA, DHA were found differently in different studies. The amounts of these fatty acids were detected low in some studies, and high in other studies. In this study, a decrease in AA and an increase in DHA (not significant) were determined. This indicates the conversion of ALA to DHA by cyclooxygenase. |
