| İlgilizce Özet | In our study, we plan to use tissue scaffolds fabricated from PLLA-PEG copolymers in order to ensure different release rates of selective growth factors. For this reason, we will synthetize PLA/PEG copolymers in the presence of stannous octoate (Sn(Oct)2) catalyst using ring-opening polymerization. Conditions of polymerization will be optimized so as to fabricate tissue scaffolds with proper , and adequate mechanical strength for utilization in the repair of mandibular defects of rabbits. After selection of the appropriate copolymer, tissue scaffolds having suitable pores with dimensions of 12 x 5 x 3 mm will be prepared by salt- extraction process. To that end, copolymer solution will be mixed up with salt particles with a certain particle-size distribution, and molded in blocks. Molded matrices will be desiccated, and salt particles within them will be removed by water- extraction method leaving a porous matrix in place of salt particles. Mechanical characteristics of the tissue scaffolds obtained will be determined by compressiveshrinkage, tensile strain and elongation tests. the need for secondary donor site, and graft material have been investigated. Porous tissue scaffolds to be prepared will be anticipated to release VEGF, and BMP2 growth factors with different release rates. Therefore, the following system design was developed. VEGF growth factor suspended within collagen or gelatin solution will be absorbed in the porous tissue scaffold. Then VEGF growth factor will be desiccated so as to form a thin layer on the porous surface, and it will be cross-linked with gaseous glutaraldehyde in order to prevent extremely rapid release of this factor when implanted in the defective areas in in vivo experiments. As for BMP2, loading of this factor in membraneous gelatin matrix prepared, and usage of this membrane as a wrapping material around tissue scaffold are planned. Thus, this membrane is expected to function as a periosteum forming a barrier between tissue scaffold, and surrounding soft tissue. This procedure is expected to free BMP2 with a release rate optimized under in vitro conditions In the next phase of the study, VEGF, and BMP2 loaded tissue scaffolds will be cultivated with a cell line similar to that of SAOS2 osteoblasts in order to determine their in vitro performances, and at certain time intervals cell formation, growth, and diffusion will be observed under scanning electron (SEM), and fluorescein microscope. |
